Mixed function oxidation. IV. An induced methylene hydroxylase in camphor oxidation.

In our earlier studies of mixed function oxidation the bicyclic monoterpene (+)-camphor (24) was used as enzyme inducer and carbon source for growth of Pseudomonas putida,1 strain Cr. The reaction sequence formulated for camphor oxidation (5) is indicated in Fig. 1: I + II + III + VII + IX ---f X with IV as a side product. A cell extract of strain C1 also forms 1,2-campholide (V) from camphor (I) (6, 7). A second organism has now been isolated which accumulates the campholide during growth on camphor. In this strain, termed Cg, camphor is metabolized by way of the second reaction sequence (8) shown in Fig. 1: I -+ V -+ VI + VII --) IX. In these pathways there are two types of mixed function oxidation, lactonization and hydroxylation. In previous reports (6, 9, 10) we have described the purification and characterization of the enzyme system catalyzing the lactonization reaction. This report describes the preliminary studies on the hydroxylation reaction, including time course of accumulation of intermediates and enzymes, as well as a formulation of the nature of the hydroxylation system.
